Effect

Lyrica is an anticonvulsant, analgesic and anxiolytic-acting gamma-aminobutyric acid analogue. It binds to voltage-dependent calcium channels, so that the channels closed, thus reducing the calcium influx at the nerve endings and the release of excitatory neurotransmitters such as glutamate, noradrenaline and substance P is decreased.

Mechanism of action

Pegabalin is a gamma-aminobutyric acid analogue with the chemical name (S) -3 - (aminomethyl)-5-methylhexanoic. It binds to a subunit of auxilia (alpha2-delta protein) of spannungsabhängigien calcium channels in the CNS and potently displaced [3H]-gabapentin. By binding the channels are closed, thus reducing the calcium influx at the nerve endings and decreases the release of excitatory neurotransmitters such as glutamate, substance P and norepinephrine. lyrica does not bind to GABA receptors and therefore has no GABAergic activity.

Absolute contraindications
- Hypersensitivity to lyrica
- Patients with rare hereditary problems of galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption

Relative contraindications
- Severe heart failure
- Pregnancy
- Breast-feeding
